What program is it you're hoping to use? What's it written in? What does it do? What OS is this on? What does it need to write to the drive? Should be possible to force a drive letter, i'll look into it.

Plug in pen drive. Right click My computer. Manage Under storage, click Disk management Right click chosen drive. Select, Change drive letter and path. Theres no way of doing it prior to plugging in the drive though afaik.
