No, open bleed when filling to let the air out as you go, leave header top off and run engine, watch and see bubbles pop up in tank. Replace cap and run engine until all hoses get hot, try heater, job done. I make it quicker by filling with hot water, this opens up the t'stat as you go.

Bleed screw is on the side of the t'stat housing, 10mm bolt.

Pull off the bottom hose and let it run, undo headatank cap as well.
