IIRC you can do it via the computer manager snap-in under drive manager. Just right click the drive you want to change and select 'change drive letters and paths' then select an empty NTFS folder to mount to.

Providing you have the basics... eg. playstation and PC networked. Just install a Upnp server and your PS3 will find it automatically. Set the Upnp server to share the video folder and you PS3 will automatically be able to stream the videos while playing. That way you don't need to copy them...
